Dosn't make sense that it stopped working. Apparently you haven't tried 'everything' because its still not working.
Check control panel sounds/microphone, make sure its turned on, test it to make sure you are getting a reading on the level meter, if this is ok, then make sure all skype settings are correct, if you can't find the problem, try a reboot, if that doesn't work, delete the application skype and reinstall it.
