"Where he knew not" means -He did not know (he was not familiar) the land where God was sending him.
Abraham was called to leave the Ur of the Chaldaeans and go to the land that God would show him. (Genesis 12-15). He obeyed God and went even though he did not know the land in which God was leading him.
