Did you lose your operating system? If it's asking for a disk, that tells me part or all of your operating system is missing.

Are you trying to add a new device? Or maybe trying to burn a CD or DVD? I'm just trying to think of reasons as to why it would ask for a disk.

Try running a virus scan. Hopefully you have a good anti virus. Maybe that will help.
What operating system are you running? (XP, Vista, Windows 7 etc...)
Is it a laptop or desktop?
Have you added any new programs recently?
How long has this been happening?
Is your DVD drive external or built into your PC?
Does your DVD tray pop open when this command pops up?
Does your PC work ok after boot up?
