Are you getting a message like "incorrect user name or password" when you attempt to login? First check for that and make sure your login details are correct. Some passwords are case sensitive so make sure your caps lock is not on.
Second, did you do anything during your last game play that may have broken the rules for game play? If so your account may be temporarily blocked. Check your email for a message about this.
Lastly, if none of these suggestions apply, check the game's help page http://s1.rail-nation.com/web/?s=1&l=com select the "I can't login option".
