A method to determine the effect of pH value on amylase activity is one that utilizes the DNS reagent (3,5-Dinitrosalicylic acid), since this reagent reacts with the products of the amylase reaction .Adition of this reagent to test tubes with different pH, in addition to carrying out a certain protocol (a specific order of actions in laboratories), the color differences between the test tubes is correlated to the enzymatic activity in them, and can be measured and quantified with spectroscopic methods.
