Why is it that while driving a car, when the windows are rolled down air only enters and doesn't leave, yet there is no build up of air pressure in the car ?

Actually when the windows are partly or fully rolled down, the laminar layer close to the surface of the car body breaks down and  the flow becomes turbulent and forms eddies or vortices. During this, air is also going out of the car and hence there is no build up of pressure.
